Did you know that compensation is all about the physics of fluorescence, and not the antibody that is being used?
Fluorescent flow cytometry is a powerful tool for understanding and characterizing complex cellular phenotypes. With the first paper in 17 color flow published in 2004, investigators and manufacturers rushed to use higher end instruments that could measure more and more fluorochromes. The fluorescent spectrum used, however, is a very crowded space and with the physics of fluorescence compounding the problem, many promising research projects are stymied because of a lack of sensitivity in the panel. In addition to using the proper controls to help interpret an experiment, proper compensation is a critical parameter to get correct. Failure to do so leads to artifacts and incorrect conclusions. In this lecture, the best practices for establishing proper compensation will be discussed. The three rules that must be followed to obtain proper compensation will be outlined, and the consequences of poor compensation will be examined.
By the end of this lecture, attendee's will understand how to recognize good and bad compensation and how to perform proper compensation quickly and easily.
