Single-Cell Mass Cytometry

Flow cytometry is an essential tool for dissecting the functional complexity of the formation of blood cellular components. Single-cell 'mass cytometry' can be used to examine many different parameters simultaneously in single cells (even up to 50 or soon even 100 different parameters like binding of antibodies, viability, DNA content, and relative cell size). By examining many parameters for each cell in populations of cells in blood, the signaling behavior of subsets of cells can be established, especially when cells are perturbed by a set of ex vivo stimuli and/or inhibitors. With these data, we are beginning to reveal the precise signaling responses of cells. Collectively, single cell analyses provide system-wide views of immune signaling that are important for understanding disease and drug response.
